The Argentine missed yet another penalty during his side's 6-0 drubbing of Getafe on Saturday, and has now missed eight spot-kicks in the Spanish top flight.
Retired Cameroon captain Eto'o previously held the record after missing seven La Liga penalties for the Catalan giants, but Messi has now overtaken him.
Overall, Barcelona's attacking trio of Luis Suarez, Neymar and Messi have already missed eight penalties in the league between them this season.
That's worse than during the Blaugrana's previous worst penalty campaign in 1976/77, when they missed seven spot-kicks, according to stats expert Mister Chip.
Messi missed four of those spot-kicks, although one of them was the penalty he passed to Suarez to score earlier in the campaign, while Neymar and the Uruguayan have each missed two in the league.
